Pertussis IgG Antibody Quantification Kit (Enzyme-Linked Immunosorbent Assay) For the quantitative detection of anti-pertussis antibodies in human serum and plasma. The reagent can be used for the diagnosis and epidemiological study of acute or recent pertussis infection, while IgG-ELISA reagent is used to monitor the vaccination.

Bordela pertussis belongs to the genus Bordela and is a small globular Gram-negative bacillus. Bordela pertussis is the causative agent of pertussis, a global infectious disease that spreads from person to person through droplets. Babies 0-4 years old are particularly vulnerable to infection. Infants and toddlers have a very high mortality rate High (Infant mortality rate within 1 year old is 60%) 1.

Bordela pertussis is attached to the mucosal ciliated cells of the respiratory tract through its surface ligands. One of the important adhesion proteins is also an important immunogen (antigen), known as Filamentous HemAgglutinin (hereinafter referred to as "FHA"). The synergistic effect of several virulence factors will promote the colonization of bacteria in the respiratory tract and trigger infection.

Bordela pertussis synthesizes and secretes a specific toxin known as Pertussis Toxin (PT), an important virulence factor. Pertussis toxin can produce complex biological effects, such as leukocytosis, lymphocytosis and the body's sensitivity to histamine increased.

A typical pertussis in three stages, after 1-2 weeks of incubation, enters the period of mucosal inflammation (highly contagious), which lasts 1-2 weeks, with a noncharacteristic cough , Rhinitis, and conjunctivitis; the next is the spasm (paroxysmal cough), paroxysmal cough often accompanied by vomiting mucus, laryngospasm and bronchospasm, resulting in infantile bruising, then repeated, sharp Of breath, then continue to cough, severe cases of seizures up to 50 times a day; 4-6 weeks later, the frequency of disease attacks will be reduced and gradually calm down (remission, the duration of up to 6 months).

Infected patients usually produce IgM-, IgA- and IgG-antibodies at the second week of the spasticity (see Figure 1), IgM-antibodies last 2-3 months, and IgA-antibodies rise from the spasm Start for 4-6 months.

IgG-antibodies peak at 6-8 weeks and are detectable until adulthood; IgM- and IgG-antibodies are produced immediay after vaccination; for infants and young children up to 6 months, specific IgM- Antibodies are suitable for the early diagnosis of recent infections, whereas specific IgA-antibodies show better diagnostic properties in infants and young children over 6 months; infants within 3 months do not produce IgA-antibodies.

【检测原理】

用抗原包被微量板孔，制成固相载体。加患者血清到板孔中，其所含的抗体特异性地与固相载体中现存抗原结合，形成免疫复合物。除去多余物质后，加入结合了碱性磷酸酶的IgG、IgA或IgM抗体，使之与上述免疫复合物反应。洗板，除去多余的结合物，加入底物（对硝基苯磷酸盐）。其与酶结合的免疫复合物反应，产生有颜色产物，颜色强度与特异性抗体含量成正比。
